Yes, you need obtain insurance on your vehicle before you register it.
According to the Colorado Division of Motor Vehicles (DMV) you must provide proof of insurance to your county clerk and recorder before registration can occur. You may use your insurance card, copy of your insurance policy or a letter from your insurance company (on the company's letterhead paper), as proof of insurance.
The CO DMV also notes that effective as of July 1, 1999, registration of your vehicle may be denied or delayed if their file does not show insurance coverage for the vehicle. Each week, insurance companies must inform the Colorado Motor Vehicle Registration Division of who has insurance and whether insurance has lapsed or does not exist.
Colorado state law requires minimum Bodily Injury Liability limits of $25,000 per injured person up to a total of $50,000 per accident, and Property Damage Liability coverage with a minimum limit of $15,000. If your car is financed though then normally your lien holder will also require that you place physical damage coverages of collision and comprehensive on your vehicle as well.
